چکیده

An experiment was conducted to immunohistochemically investigate myogenesis and the expression pattern of two myogenic regulatory proteins, myf-3 and myogenin, during formation of secondary fibres in pigs. Muscle fibres develop as two distinct populations in the foetal pig. Our data in foetal pigs suggest that formation of secondary fibres occurs from about day 60 to 80 of gestation and that myogenesis is concluded at approximately day 80 of gestation. The frequency of nuclei expressing myf-3 did not change, while that of nuclei expressing myogenin decreased from day 60 to 100 of gestation. Thus, myf-3 and myogenin appear to exhibit different expression patterns during myogenesis. The significance of decline in the frequency of nuclei expressing myogenin during secondary muscle fibre formation is not clear. During postnatal muscle growth no clear relationships between the expression pattern of myf-3 and myogenin with different fibre types were established.  2000 Elsevier Science B.V. A unique pattern of expression of the four muscle regulatory factor proteins distinguishes somitic from embryonic, fetal and newborn mouse myogenic cells.

A unique pattern of expression of the four muscle regulatory factor (MRF) proteins was found to distinguish early somitic from embryonic, fetal and newborn limb myogenic cells in vitro. Expression of the myosin heavy chain (MHC), MyoD, myogenin, Myf-5, and MRF4 proteins was examined by immunocytochemistry in cultures of four distinct types of mouse myogenic cells: somitic (E8.5), embryonic (E11...

Pax3 induces differentiation of juvenile skeletal muscle stem cells without transcriptional upregulation of canonical myogenic regulatory factors.

Pax3 is an essential myogenic regulator of fetal and embryonic development, but its role in postnatal myogenesis remains a topic of debate. We show that constitutive expression of Pax3 in postnatal, juvenile mouse skeletal muscle stem cells, a subset of the heterogeneous satellite cell pool highly enriched for myogenic activity, potently induces differentiation. This differentiation-promoting a...

Ectopic Pax-3 Activates MyoD and Myf-5 Expression in Embryonic Mesoderm and Neural Tissue

To understand how the skeletal muscle lineage is induced during vertebrate embryogenesis, we have sought to identify the regulatory molecules that mediate induction of the myogenic regulatory factors MyoD and Myf-5. In this work, we demonstrate that either signals from the overlying ectoderm or Wnt and Sonic hedgehog signals can induce somitic expression of the paired box transcription factors,...

Repression through a distal TCF-3 binding site restricts Xenopusmyf-5 expression in gastrula mesoderm

The development of skeletal muscle in the vertebrate embryo is controlled by a transcriptional cascade that includes the four myogenic regulatory factors Myf-5, MyoD, Myogenin, and MRF4. The dynamic expression pattern of myf-5 during myogenesis is thought to be consistent with its role during early determination of the myogenic lineage. To study the factors and mechanisms, which regulate myf-5 ...

Arginine-vasopressin induces differentiation of skeletal myogenic cells and up-regulation of myogenin and Myf-5.

The neurohypophyseal nonapeptide arginine8-vasopressin (AVP) induces phosphoinositide turnover and calcium and pH changes in skeletal myogenic cells in culture. In order to investigate the effect of AVP on skeletal myogenesis, we examined the effect of this hormone on proliferating mononucleated L6 myoblast cultures. Addition of AVP to the medium resulted in the formation of much larger myotube...
